麻醉和手术对5XFAD转基因小鼠海马相关蛋白表达影响及其性别差异的研究

Effects of anesthesia surgery on the expression of hippocampal related proteins in 5XFAD transgenic mice and their gender differences

  • 摘要: 目的 探究麻醉和手术是否影响5XFAD (Transgenic Mice with Five Familial Alzheimer’s Disease Mutations)转基因小鼠海马中相关蛋白的表达并研究有无性别差异。方法 将5XFAD转基因小鼠与C57BL/6J野生型(Wild-Type, WT)小鼠进行合笼繁殖得到子代小鼠并进行基因鉴定。将4月龄子代5XFAD转基因小鼠和同窝同龄阴性(Littermate,LM)小鼠,按雌雄性别选取各32只,进而将每种性别小鼠各分为4组:5XFAD对照组(5XFAD Control组)、5XFAD麻醉/手术处理组(5XFAD Anesthesia/surgery组)、 LM对照组(LM Control组)和LM麻醉/手术处理组(LM Anesthesia/surgery组),每组8只。将随机进入麻醉和手术处理组的实验小鼠在1.4%异氟醚麻醉下进行开腹手术处理,对照组不行麻醉和手术,于术后24h取海马组织,采用Western Blot法检测β-catenin、糖原合成酶激酶(glycogen synthase kinase 3β, GSK3β)及p-GSK3β蛋白含量。结果 在雌性5XFAD鼠中,与5XFAD Control组相比,5XFAD Anesthesia/surgery组海马组织中β-catenin、p-GSK3β含量及其p-GSK3β/GSK3β比值减少(P<0.05);在雄性5XFAD鼠中,5XFAD Control组与5XFAD相比,β-catenin、p-GSK3β含量及其p-GSK3β/GSK3β比值无统计学意义(P>0.05);与雄性5XFAD Anesthesia/surgery 组相比,雌性5XFAD Anesthesia/surgery组β-catenin、p-GSK3β含量及其p-GSK3β/GSK3β比值减少(P<0.05);在雌性和雄性LM鼠中,与各自LM Control组比较,LM Anesthesia/surgery组β-catenin、p-GSK3β含量及其p-GSK3β/GSK3β比值无统计学意义(P>0.05)。结论 麻醉和手术后5XFAD转基因小鼠海马中β-catenin蛋白表达水平以及p-GSK3β/GSK3β比值降低,其中雌性更易感。

     

    Abstract: []Objective To investigate whether anesthesia and sursgery affect the expression of related proteins in the hippocampus of 5XFAD (Transgenic Mice with Five Familial Alzheimer’s Disease Mutations) transgenic mice and to study whether there are gender differences. Methods 5XFAD transgenic mice were bred with C57BL/6J wild-type (WT) mice to obtain offspring mice and perform gene identification. 4-month-old offspring 5XFAD transgenic mice and littermate (LM) mice of the same age were selected, 32 males and 32 females, and then divided into 4 groups of each sex: 5XFAD control group, 5XFAD anesthesia/surgery group, LM control group and LM anesthesia/surgery grou, with 8 mice in each group. The experimental mice randomly assigned to the anesthesia and surgery groups underwent laparotomy under 1.4% isoflurane anesthesia, while the control group did not undergo anesthesia or surgery.
