Wnt/β-catenin信号通路在眼部病理性血管新生过程中作用的研究进展

Research progress of Wnt/β-catenin signaling pathway regulating ocular pathological angiogenesis

  • 摘要: 眼部病理性新生血管相关疾病会对视力造成显著影响,目前临床一线治疗方案为眼内注射抗血管内皮生长因子类药物,但存在应答欠佳、潜在不良反应等局限性。Wnt/β-catenin信号通路有促进胚胎发育、维持组织稳态的功能,在组织中广泛表达。Wnt/β-catenin信号通路异常激活时,下游基因产物的表达增加能够上调缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)等细胞因子,促进眼部的病理性血管新生。Wnt抑制剂能够延缓眼部病理性血管新生进展。Wnt/β-catenin信号通路与眼部血管新生病理机制的研究为眼部病理性新生血管相关疾病的治疗提供了新的潜在靶点。

     

    Abstract: Ocular pathological neovascularization diseases can significantly impair patients' visual acuity. At present, intraocular injection of anti-vascular endothelial growth factor (anti-VEGF) drugs is widely used as the first-line treatment, but there are limitations such as poor response and potential adverse reactions. Wnt/β-catenin signaling pathway has the function of promoting embryonic development and maintaining tissue homeostasis, and is widely expressed in tissues in vivo. When the Wnt/β-catenin signaling pathway is abnormally activated, the expression of downstream gene products can upregulate cytokines like hypoxia-inducible factor-1α (HIF-1α), and then promote pathological neovascularization in the eye. The application of Wnt inhibitors can delay the progression of pathological angiogenesis in the eye. The study of Wnt/β-catenin signaling pathway and pathological mechanism or ocular angiogenesis provides new potential targets for the treatment of ocular pathologic neovascularization diseases.
