IL-1β基因多态性与儿童紫癜性肾炎易感性的相关性分析

Association between IL-1β gene polymorphism and susceptibility to Henoch-Schönlein purpura nephritis in children

    目的 探讨白细胞介素1β(IL-1β)基因单核苷酸多态性与儿童过敏性紫癜及紫癜性肾炎易感性的关系。
    方法 选择过敏性紫癜患儿174例作为病例组,将同期体检的162例健康儿童作为对照组。根据临床症状,将病例组患儿分为单纯型组、关节型组、腹型组、混合型组;根据病例组患儿在随访过程中是否合并肾脏损害分为非紫癜性肾炎组、紫癜性肾炎组。采用M-PCR法靶向捕获IL-1β基因rs1143627位点,通过高通量测序技术对所有样本进行该位点测序,并对测序数据与不同分组进行关联性分析。
    结果 rs1143627基因型及等位基因在各临床症状分组中差异无统计学意义;rs1143627基因型GG及等位基因G在病例组与对照组、非紫癜性肾炎组与紫癜性肾炎组间差异均有统计学意义(P < 0.05)。
    结论 IL-1β基因rs1143627位点多态性与儿童过敏性紫癜的发生及肾脏受累有关;等位基因G可能是儿童过敏性紫癜的易感基因,且与紫癜性肾炎的发生有关。

     

    Abstract:
    Objective To investigate the relationship between single nucleotide polymorphism of interleukin-1β (IL-1β) gene and susceptibility to Henoch-Schönlein purpura and Henoch-Schönlein purpura nephritis in children.
    Methods A total of 174 children with Henoch-Schönlein purpura were selected as the case group, and 162 healthy children who underwent physical examination at the same time were selected as the control group. According to their clinical symptoms, patients were divided into the simple group, joint group, abdominal group, and mixed groups. According to whether complicated with renal damage during the follow-up, patients were divided into non-nephritis group and nephritis group. The rs1143627 locus of the IL-1β gene was targeted and captured by the M-PCR method. All samples were sequenced by the high-throughput sequencing technique, and the correlations of the sequencing data and the different groups were analyzed.
    Results There was no significant difference in rs1143627 genotype and allele among different clinical symptom groups, but there were significant differences in rs1143627 genotype GG and allele G between the case group and control group, and the non-nephritis group and nephritis group.
    Conclusions rs1143627 gene polymorphism is associated with Henoch-Schönlein purpura and renal involvement. Allele G may be the susceptible gene of Henoch-Schönlein purpura and is related to the occurrence of Henoch-Schönle purpura nephritis in children.
