Abstract:
Objective:To investigate the inhibitory effect of retinoic-acid-related orphan nuclear receptor C (RORC/RORγ) on the proliferation of Siha and HeLa cells and explore the possible mechanisms. Methods:The inhibitory effect of RORC on the proliferation of Siha and HeLa cells was evaluated by cell counting kit-8 (CCK8) assay. Glucose levels in Siha and HeLa cells were measured using glucose assay kit. The expression levels of RORC, glucose transporter 1 (GLUT1), lactate dehydrogenase A (LDHA), and hypoxia-inducible factor 1α (HIF-1α) were determined by Western blotting. Results:The results of CCK-8 showed that RORC significantly inhibited the proliferation of Siha and HeLa cells (P<0.05). Glucose metabolism test results showed that glucose uptake in RORC overexpression cells decreased (P<0.05). Western blotting results showed that the expressions of GLUT1, LDHA, and HIF-1α in RORC overexpression cells were downregulated markedly (P<0.05). Conclusions:RORC could inhibit the proliferation of the Siha and HeLa cervical cancer cell lines through inhibition of their glucose metabolism.
