维甲酸相关孤儿核受体C可通过抑制糖酵解来调节宫颈癌细胞增殖

Retinoic-acid-related orphan nuclear receptor C regulates cervical cancer cell proliferation by inhibiting glycolysis

  • 摘要: 目的:探讨维甲酸相关孤儿核受体C(RORC/RORγ)对宫颈癌细胞Siha和HeLa的增殖抑制作用及相关机制。方法:采用CCK-8(cellcountingkit-8)法检测RORC对宫颈癌细胞增殖的抑制作用;采用葡萄糖检测分析试剂盒进行细胞内糖代谢检测分析;采用蛋白质印迹法(Western印迹)检测RORC、葡萄糖转运因子1(GLUT1)、乳酸脱氢酶A(LDHA)和缺氧诱导因子1α(HIF-1α)表达的变化。结果:CCK-8检测结果显示,RORC能明显抑制宫颈癌细胞Siha和HeLa细胞增殖(P<0.05)。糖代谢检测结果显示,RORC能使宫颈癌细胞葡萄糖摄取水平降低(P<0.05)。Western印迹检测结果显示,RORC可使宫颈癌细胞内GLUT1、LDHA和HIF-1α蛋白表达水平下调(P<0.05)。结论:RORC可通过降低宫颈癌细胞Siha和HeLa糖代谢水平来抑制细胞增殖。

     

    Abstract: Objective:To investigate the inhibitory effect of retinoic-acid-related orphan nuclear receptor C (RORC/RORγ) on the proliferation of Siha and HeLa cells and explore the possible mechanisms. Methods:The inhibitory effect of RORC on the proliferation of Siha and HeLa cells was evaluated by cell counting kit-8 (CCK8) assay. Glucose levels in Siha and HeLa cells were measured using glucose assay kit. The expression levels of RORC, glucose transporter 1 (GLUT1), lactate dehydrogenase A (LDHA), and hypoxia-inducible factor 1α (HIF-1α) were determined by Western blotting. Results:The results of CCK-8 showed that RORC significantly inhibited the proliferation of Siha and HeLa cells (P<0.05). Glucose metabolism test results showed that glucose uptake in RORC overexpression cells decreased (P<0.05). Western blotting results showed that the expressions of GLUT1, LDHA, and HIF-1α in RORC overexpression cells were downregulated markedly (P<0.05). Conclusions:RORC could inhibit the proliferation of the Siha and HeLa cervical cancer cell lines through inhibition of their glucose metabolism.
