悬浮球培养法制备ABCG2阳性表达胃癌细胞株

Preparation of ABCG2 positive gastric cancer cell line by spheroid cell suspension culture method

  • 摘要: 目的:探讨悬浮球培养法制备具有干细胞特性(ABCG2阳性表达)胃癌细胞株MKN45的可行性。方法:采用悬浮球培养法在含成纤维细胞生长因子(bFGF)及表皮生长因子(EGF)的无血清培养基中培养人胃癌细胞株MKN45,观察细胞球体的形成过程,并检测球体细胞及原代贴壁细胞中干细胞标志物ABCG2的表达水平。结果:在无血清培养基培养条件下,胃癌细胞MKN45能形成悬浮球体;球体细胞ABCG2表达水平高于原代贴壁细胞,差异有统计学意义(P<0.05)。结论:悬浮球培养法可制备具有肿瘤干细胞特性的ABCG2阳性表达胃癌细胞株,为后续研究奠定了基础。

     

    Abstract: Objective:To investigate the feasibility of preparing ABCG2 positive gastric cancer cell line MKN45 by spheroid cell suspension culture method. Methods:The human gastric cancer cells MKN45 were cultured in the serum free medium which contained fibroblast growth factor (bFGF) and epidermal growth factor (EGF). The formation of spheroid was investigated and the expression levels of stem cell marker ABCG2 in the spheroid cells and primary adherent cells were detected. Results:Under the condition of serum free culture, the gastric cancer cell line MKN45 could form spheroid. The expression level of ABCG2 in the spheroid cells was higher than that in the parental cells, the difference was statistically significant (P<0.05). Conclusions:The spheroid cells formed by suspension culture of gastric cancer cell line MKN45 possess the characteristics of cancer stem cells.
